Penelitian ini bertujuan mempelajari farmakokinetik hidroflumetiazida (HFMT; abat model) pada keadaan gagal ginjal akut karena uranil nitrat.
Setelah pemberian uranil nitrat Oasis tunggal (15 mg/kg;ip) kepada tikus, terjadi pengurangan volume urin dan jumlah kreatinin urin yang diekskresi, hal mana peristiwa ini menunjukkan gejala kegagalan ginjal tikus.
Farmakokinetik HFMT (dosis tunggal 10 mg/kg; ip) pada tikus gagal ginjal térnyata berbeda dengan farmakokinetik obat itu pada tikus normal, kecuali tetapan kecepatan absorpsi (Ka). Kadar HFMT maksimum (Cmaks) dalam darah naik 2,5-kalinya, sedangkan waktu untuk mencapai Cmaks tertunda hampir dart normal. Nilai volume distribusi HFMT berkurang hampir 2-kalinya dibandingkan dengan nilainya pada ginjal normal, yakni dart 112 ml/kg sampai 60,9 - 65,1 ml/kg (pc0,05). Klirens HFMT berkurang (antara 1,5-2 kalinya) dart normal, sehingga memperpanjang waktu-paro eliminasinya (pc0,05), dart 41 menit sampai berkisar antara 319 -512 menit Harga AUC HFMT pada tikus gaga/ ginjal naik antara 14-21 kali dart normal (pc0,05).
ABSTRACT
This investigation was performed to study the pharmacokinetics of a model drug hydroflumethiazide (HFMT) in rats with acute renal dysfunction due to urany! nitrate.
Following single dose administration of urany! nitrate (15 mg/kg; ip) to the animals, a gradual decrease of urine volume and urinary creatinine excreted has occurred from day to day indicating the existence of renal dysfunction.
Pharmacokinetics of HFMT (single dose 10 mg/kg; ip) in rats with renal failure differ to those in normal rats, except the absorption rate constant (Ka). Maximum HFMT concentration (Cmax) in blood increased 2.5-fold, whereas the time to reach Cmax delayed by almost twice of that normal value. Volume of distribution of HFMT decreased by two-fold compared to that in normal, i.e. from 112 ml/kg to 60.9-65.1 ml/kg (p,(0.05). Clearance value of the drug also decreased by about 1.5-2 fold, which in turn prolonged the elimination half-life (pc0.05) from 41 minutes to 319-512 minutes.
